O DNA \u00e9 o reposit\u00f3rio central da informa\u00e7\u00e3o gen\u00e9tica e n\u00e3o \u00e9 diretamente utilizado para produzir prote\u00ednas. A express\u00e3o g\u00eanica come\u00e7a com a transcri\u00e7\u00e3o, quando uma c\u00f3pia do DNA que comp\u00f5e o gene \u00e9 feita ou transcrita em um RNA mensageiro precursor (pr\u00e9-mRNA). O pr\u00e9-mRNA \u00e9 composto por exons e regi\u00f5es intervenientes n\u00e3o codificantes chamadas introns. Os introns s\u00e3o removidos \u2013 ou \u201cemendados\u201d \u2013 e os exons s\u00e3o unidos em uma ordem espec\u00edfica. Este RNA mensageiro maduro inclui todas as informa\u00e7\u00f5es necess\u00e1rias para produzir prote\u00ednas funcionais.&nbsp;<\/p>\n\n\n\n<h4 class=\"wp-block-heading\" id=\"h-splicing-alternativo-nbsp\">Splicing alternativo&nbsp;<\/h4>\n\n\n\n<p class=\"core-block core-paragraph\">Um \u00fanico gene pode codificar mais de um tipo de prote\u00edna. Isso pode ocorrer durante um processo chamado splicing alternativo, em que diferentes conjuntos de exons podem ser unidos a partir de um gene, criando sequ\u00eancias codificadoras alternativas e m\u00faltiplas. Essas sequ\u00eancias geram m\u00faltiplos produtos de RNA mensageiro que codificam prote\u00ednas diferentes e relacionadas. Aproximadamente 94% de todos os genes humanos passam por splicing alternativo como parte da produ\u00e7\u00e3o de prote\u00ednas funcionais.&nbsp;<\/p>\n\n\n\n<p class=\"core-block core-paragraph\">Uma m\u00e1quina molecular complexa chamada spliceossomo \u00e9 a principal respons\u00e1vel pelo splicing. O spliceossomo \u00e9 composto por cinco pequenos RNPs nucleares (snRNPs), que se organizam em torno dos locais de splicing que servem como limites entre cada exon e intron no pr\u00e9-mRNA. O primeiro passo na montagem do spliceossomo \u00e9 mediado pelos snRNPs U1 e U2. A intera\u00e7\u00e3o U1\u2013pr\u00e9-mRNA \u00e9 a mais extensa e seletiva na via de splicing. Uma vez montado, o spliceossomo catalisa a remo\u00e7\u00e3o dos introns, seguida pela uni\u00e3o dos exons conforme necess\u00e1rio.&nbsp;<\/p>\n\n\n\n<h4 class=\"wp-block-heading\" id=\"h-mutacao-no-splicing-nbsp\">Muta\u00e7\u00e3o no splicing&nbsp;<\/h4>\n\n\n\n<p class=\"core-block core-paragraph\">Muta\u00e7\u00f5es ou diversidade de sequ\u00eancias que interrompem, enfraquecem ou ativam alternadamente os locais de splicing podem afetar a capacidade dos componentes do spliceossomo de reconhecer com precis\u00e3o o pr\u00e9-mRNA. Essas muta\u00e7\u00f5es s\u00e3o a causa direta de mais de 200 doen\u00e7as humanas, incluindo muitos tipos de c\u00e2ncer e doen\u00e7as gen\u00e9ticas, como atrofia muscular espinhal e doen\u00e7a de Huntington. Em muitos casos, erros de splicing resultam na falha da intera\u00e7\u00e3o correta entre o U1 snRNP e o pr\u00e9-mRNA devido \u00e0 diversidade estrutural no local de splicing. Essas mudan\u00e7as resultam na altera\u00e7\u00e3o das efici\u00eancias de splicing.&nbsp;<\/p>\n\n\n\n<h4 class=\"wp-block-heading\" id=\"h-modificadores-de-splicing-nbsp\">Modificadores de splicing&nbsp;<\/h4>\n\n\n\n<p class=\"core-block core-paragraph\">A PTC desenvolveu uma tecnologia poderosa que permite a identifica\u00e7\u00e3o de pequenas mol\u00e9culas conhecidas como modificadores de splicing \u2013 projetadas para modificar a intera\u00e7\u00e3o U1\u2013pr\u00e9-mRNA. Esses diferentes tipos de mol\u00e9culas funcionam como uma \u201ccola\u201d molecular para fortalecer a intera\u00e7\u00e3o no local de splicing e ajudar a tornar o splicing mais eficiente.&nbsp;<\/p>\n\n\n\n<p class=\"core-block core-paragraph\">A PTC foi pioneira na no\u00e7\u00e3o de que o splicing pode ser modulado utilizando pequenas mol\u00e9culas. Nos \u00faltimos 20 anos, aprimoramos nossa tecnologia de splicing de RNA. Usando essa tecnologia, identificamos com sucesso uma mol\u00e9cula oral para o tratamento da atrofia muscular espinhal, que foi a primeira prote\u00edna de mol\u00e9cula pequena aprovada para intervir no splicing, o que levou \u00e0 produ\u00e7\u00e3o de uma prote\u00edna SMN est\u00e1vel e funcional.\u00a0<\/p>\n\n\n\n<p class=\"core-block core-paragraph\">Al\u00e9m disso, temos v\u00e1rios programas em descoberta e desenvolvimento usando nossa plataforma de splicing. Nosso pr\u00f3ximo composto mais avan\u00e7ado est\u00e1 sendo desenvolvido para o tratamento da doen\u00e7a de Huntington. Essa doen\u00e7a rara \u00e9 causada por um gene defeituoso que produz uma prote\u00edna mutada chamada HTT.&nbsp;&nbsp;<\/p>\n\n\n\n<p class=\"core-block core-paragraph\">A tecnologia de splicing alternativo de RNA tem um amplo potencial para identificar novos tratamentos para muitas doen\u00e7as.
